Each mission features different squadmates that provide support to player in each situation. The player takes control of Alpha Team leader Domingo Chavez on each mission and with either John Clark or Alistair Stanley serving as onsite coordinators. The international terrorist organization Global Liberation Front has stolen a nanotech aerosol virus known as Legion. To emphasize the more arcade combat, enemy accuracy has been significantly reduced. Despite this, players are unable to restore their health during a mission.

Both the player and enemies are now more durable and are able to take several hits before dying. Squadmate AI has been improved, allowing them to perform more independent actions. Like past titles, the player issues orders to AI controlled teammates such as breaching rooms, tossing grenades, etc. Lockdown's gameplay is more action-oriented then that of past installments which focused more on realism and tactics.
